Simply put, yes if you exceed the "Daily API Requests per App" limit in Kintone, you will most likely experience interruptions in your application's functionality. I'm not 100% sure so you should contact the Kintone support desk if you would like confirmation. However, most likely any API requests will be rejected until the limit resets the following day.
Just so you are aware, the maximum number of API requests available per day per app is 10,000 requests.

To my knowledge, there is is no way to increase the maximum limit of API requests. You could check with the Kintone support desk to see if they have any additional information.
